The presence of PI drugs can block the ability of viral protease enzymes from cleaving non-functional viral protein precursors into their functional subunits. This animation describes how mutations in viral proteases can lead to resistance to PI drugs. As a consequence of drug resistance, viral proteases can continue to cleave non-functional protein precursors into functional subunits, permitting the assembly of new viral particles.
